Evidence for Two Classes of Prereplicative Cistrons*

چکیده
The antibiotic rifampicin has been used in an investigation of the temporal regulation of gene expression during bacteriophage T4 infection of Escherichia coli. We have asked at what times after infection the transcription of specific cistrons becomes rifampicin insensitive. Gene expression has been studied using analyses of proteins on sodium dodecyl sulfate-acrylamide gels; thus translation has been used to assay for the appearance of specific transcripts. Two types of promoters account for most prereplicative transcription; early promoters are recognized immediately after infection, whereas quasi-late promoters are recognized only after a delay of 14 min. Those transcripts previously defined as immediate early and delayed early RNAs initially appear to be under the control of early promoters; however, the quasi-late promoters may provide a second mode of regulation for several of those transcripts.

متن کاملTranscription and translation of prereplicative bacteriophage T4 genes in vitro.
Bacteriophage T4 DNA was used to direct transcription and translation in vitro in extracts prepared from uninfected Escherichia coli. The radioactive protein products of the cell-free reactions were examined on sodium dodecyl sulfate acrylamide gels. We conclude that the cell-free system prepared from uninfected E. coli has the capacity to synthesize most T4 prereplicative RNAs and proteins. Fu...
متن کاملEvidence for position-effect suppression of the ribosomal RNA cistrons in Drosophila melanogaster.
Unambiguous evidence is presented that two X-chromosome inversions produce lethality in X/O males because of position-effect suppression of genes in the basal heterochromatin. The data support the hypothesis that the ribosomal RNA cistrons are the genes suppressed. There is evidence for a region in the X basal heterochromatin that may act as a regulator of rDNA activity.
متن کاملCharacterization of cytoplasmic and nuclear genomes in the colorless alga Polytoma. III. Ribosomal RNA cistrons of the nucleus and leucoplast
The colorless alga Polytoma obtusum has been found to possess leucoplasts, and two kinds of ribosomes with sedimentation values of 73S and 79S. The ribosomal RNA (rRNA) of the 73S but not the 79S ribosomes was shown to hybridize with the leucoplast DNA (rho - 1.682 g/ml). Nuclear DNA of Polytoma (rho = 1.711) showed specific hybridization with rRNA from the 79S ribosomes. Saturation hybridizati...
متن کاملC-terminal region of herpes simplex virus ICP8 protein needed for intranuclear localization.
The herpes simplex virus single-stranded DNA-binding protein, ICP8, localizes initially to structures in the nucleus called prereplicative sites. As replication proceeds, these sites mature into large globular structures called replication compartments. The details of what signals or proteins are involved in the redistribution of viral and cellular proteins within the nucleus between prereplica...
